Molecular markers based genetic relatedness studies in tissue culture propagated japanese plum cultivars Santa Rosa and Frontier

Santa Rosa and Frontier are the major Japanese plum (Prunus salicina Lindl.) cultivars grown throughout world. The present investigation was performed to understand genetic relatedness among in vitro propagated using PCR based molecular markers. For study, three arbitrary markers viz. RAPD (Random amplified Polymorphic DNA), ISSR (Inter-Simple Sequence Repeats) SCoT (Start Codon Targeted) were used. In analysis, 18 primers out of 28 generated 33 scorable bands. allelic variations when analysed, revealed 84% similarity between these two with highest polymorphic information content 0.78. Similarly, 15 produced 73 amplicons an average 4.86 amplicon per primer coefficient ranging from 62 67%. Seven 26 resulted a total twenty- six bands 24 Cluster analysis all used broadly divided into clusters containing shoots, their progenies mother trees respective genotypes. study concluded that marker systems found be effective revealing relationship commercially important cultivars.
